TLX3 also known as T-cell leukemia homeobox protein 3 is a transcription factor with a mass of approximately 33 kDa. It belongs to the homeobox gene family and plays a role in regulating gene expression. TLX3 shows expression mostly in the nervous system and immature T-cells particularly during development. The protein acts by binding to specific DNA sequences leading to activation or suppression of target genes essential for the control of developmental processes.
TLX3 influences the fate of neuronal cell types and is critical in determining neurotransmitter identity. It functions as part of a regulatory complex that guides the differentiation of neurons especially in the dorsal spinal cord. Its activity impacts pathways that promote the production of excitatory glutamatergic neurons over inhibitory GABAergic neurons ensuring proper balance in the central nervous system.
TLX3 operates in the gene regulatory networks connected to neuronal differentiation and survival. It interacts importantly with the Notch signaling pathway influencing neurogenesis and cell fate decisions. TLX3 shares regulatory pathways with proteins such as TLX1 and influences the expression of downstream genes that are required for neuronal identity and function. TLX3 is linked to T-cell acute lymphoblastic leukemia (T-ALL) and has implications in certain neurodevelopmental disorders.
