TM6SF2 also known as Transmembrane 6 Superfamily Member 2 is a protein involved in lipid metabolism. It has a mass of approximately 42 kDa. TM6SF2 expresses mainly in the liver and to a lesser extent in the small intestine. It plays a mechanical role in modulating triglyceride metabolism by affecting very-low-density lipoprotein (VLDL) secretion from the liver. TM6SF2 influences the steady-state levels of triglycerides in hepatocytes by promoting the degradation of triglycerides within the endoplasmic reticulum.
Biological function summary

TM6SF2 impacts lipid and energy homeostasis. It does not form part of a large protein complex but it has an independent role in cellular metabolism. The modulation of VLDL secretion by TM6SF2 helps maintain appropriate lipid levels in circulation contributing to overall metabolic health. It exhibits structural aspects that allow it to interact with other molecules within the cellular lipid landscape influencing cholesterol homeostasis as well.

Pathways

TM6SF2 is involved in the lipid metabolism and energy homeostasis pathways. It interacts with other proteins like APOE and LPCAT1 to regulate lipid levels within these pathways. TM6SF2 acts as an important modulator by affecting lipid processing and sequestration. The regulation of these pathways ensures balanced lipid concentrations which is essential for normal cellular and systemic functions.

TM6SF2 associates with non-alcoholic fatty liver disease (NAFLD) and cardiovascular disease. A mutation in TM6SF2 can increase susceptibility to NAFLD by causing inefficient hepatic lipid processing. This mutation known as E167K links TM6SF2 to accumulation of fat in the liver. TM6SF2 also connects with proteins like APOB through the influence on lipid levels contributing to cardiovascular risk. Understanding its role offers potential for targeting these diseases therapeutically.
