TMC3 also known as Transmembrane Channel-like 3 is a protein encoded by the TMC3 gene in humans. It has a molecular mass of approximately 110 kDa. This protein is part of the transmembrane channel-like family and is primarily expressed in the cochlea and vestibular system of the inner ear. TMC3 is believed to act as a cation channel which is important for the sensory function of both hearing and balance in mammals. The structural configuration allows it to facilitate ion transport across cell membranes.
Biological function summary

TMC3 functions as a component in mechanotransduction processes in the inner ear. As part of the transmembrane channel complex it works collaboratively with other proteins like TMC1 and TMC2 to mediate mechanosensory transduction in hair cells. This activity is essential for converting mechanical stimuli into electrical signals which then travel to the brain as part of auditory and vestibular processing. Though not as well studied as TMC1 the role of TMC3 in this complex suggests it might have stabilizing or supportive functions in the transduction pathway.

Pathways

TMC3 is involved in the mechanosensory signaling pathway within the inner ear. It interacts with other proteins like TMC1 and TMC2 in this pathway to facilitate auditory and balance perception. The pathway includes calcium signaling critical for maintaining hair cell function and communication with the nervous system. TMC3 is not only distinguished by these associations but it also plays a part in synaptic activity important for hearing and balance.

TMC3's disruption may contribute to conditions like hearing loss and vestibular dysfunction. Genetic variations in TMC3 and related proteins such as TMC1 and TMC2 are linked to hereditary forms of hearing impairment. Although research is still ongoing evidence suggests that altered TMC3 expression can impair mechanotransduction leading to sensorineural hearing issues. Understanding TMC3's role could provide insight into therapeutic targets for treating such auditory disorders.
