Supplementary information

TMEM135 also known as Transmembrane Protein 135 has a molecular mass of approximately 50 kDa. This protein is expressed in multiple tissues with high levels found in adipose tissue and heart. It functions mechanically by regulating lipid metabolism and its activity is associated with mitochondria. TMEM135 helps maintain mitochondrial dynamics including fusion and fission processes which are critical for proper energy homeostasis in cells.
Biological function summary

TMEM135 participates in energy balance and lipid metabolism. This protein does not function alone; it is part of complexes involving mitochondrial membranes. Within these complexes TMEM135 helps orchestrate the metabolism of lipids impacting overall energy distribution in cells. Its role extends to regulating oxidative stress suggesting it plays a part in cellular responses to metabolic changes.

Pathways

TMEM135 functions contribute to the mitochondrial apoptosis pathway and the peroxisome proliferator-activated receptor (PPAR) signaling pathway. These pathways are essential in controlling cell viability and energy metabolism respectively. TMEM135 interacts with proteins like DRP1 and PGC-1α which further help in mitochondrial division and energy metabolism regulation reflecting its importance in cellular energetics.

TMEM135 displays implications in metabolic syndrome and cardiovascular disease. Its influence on lipid metabolism and mitochondrial function links it to these conditions. In metabolic syndrome altered function of TMEM135 may affect PGC-1α involved in the regulation of glucose and lipid homeostasis. In cardiovascular disease disrupted mitochondrial dynamics due to TMEM135 dysfunction can lead DRP1-driven imbalances resulting in compromised heart function.

Target data

Involved in mitochondrial metabolism by regulating the balance between mitochondrial fusion and fission. May act as a regulator of mitochondrial fission that promotes DNM1L-dependent fission through activation of DNM1L. May be involved in peroxisome organization.
