Tmem18 also known as Transmembrane Protein 18 is a transmembrane protein with an approximate mass of 17 kDa. It is involved in cellular regulation through its action on chromatin and DNA binding impacting gene expression. Tmem18 is mainly expressed in the brain particularly in the hypothalamus and to a lesser extent in other tissues like adipose (fat) tissue. Current research focuses on the Tmem18's role in neural cell migration and its influence on cellular signaling pathways.
Biological function summary

The protein impacts neural development and energy homeostasis through modulation of transcriptional activities. Tmem18 does not belong to a well-defined complex but it associates with cell surface receptors playing a role in cellular communication and signaling transduction. Through these interactions it alters the expression of genes that control key functions in brain development and energy metabolism notably influencing feeding behavior and energy balance in organisms.

Pathways

Tmem18 associates with both the Wnt signaling pathway and the hypothalamic signaling involved in energy regulation. Its interaction with these pathways influences cellular mechanisms like neural proliferation and differentiation. The protein collaborates with other proteins in these pathways such as beta-catenin in the Wnt pathway and neuropeptide Y in the hypothalamic regulation of appetite highlighting its contribution to cellular and physiological balance.

Tmem18 links to obesity and neurodegenerative conditions. Alterations in Tmem18 expression or function relate to abnormal energy homeostasis contributing to obesity. Furthermore dysregulation in its expression affects neural cells associating it with neurodegenerative diseases. Tmem18 interacts with other proteins involved in such conditions including leptin in the context of obesity and tau proteins in neurodegenerative disorders illustrating its significant role in these areas.
