Supplementary information

TMEM24 also known as transmembrane protein 24 is an integral membrane protein with approximately 56 kDa mass. It localizes primarily in the endoplasmic reticulum (ER) and is present in a variety of tissues including the brain and pancreas. It plays a role in maintaining lipid homeostasis acting as a lipid transfer protein with the ability to shuttle phosphatidylinositol from the ER to the plasma membrane. TMEM24 has regions that facilitate membrane association and phospholipid transfer which are key for its mechanical function.
Biological function summary

TMEM24 contributes to cellular processes by regulating glucose-stimulated insulin secretion. It does this by forming part of a complex that modulates the availability of membrane lipids necessary for signaling and membrane dynamics. The protein's activity affects the response of pancreatic beta cells influencing insulin release making it an important player in metabolic regulation. TMEM24 also interacts with other proteins involved in lipid metabolism making it essential for energy homeostasis.

Pathways

TMEM24 is important in the insulin signaling and glucose metabolism pathways. It coordinates with proteins such as PI4KIIIβ and E-syts which are important for phosphoinositide lipid synthesis and turnover. These interactions ensure efficient lipid signaling and membrane trafficking contributing to the proper functioning of insulin signaling. TMEM24's role in these pathways underlines its importance in managing cellular lipid levels and energy balance.

TMEM24 has been implicated in metabolic disorders like type 2 diabetes. Its functional interaction with proteins involved in lipid signaling and insulin secretion such as INS and IRS-1 points to its involvement in glucose homeostasis and pancreatic beta-cell response. Alterations in TMEM24 expression or function may lead to impaired insulin secretion and insulin resistance contributing to diabetes pathogenesis. Understanding TMEM24's role in these processes may offer insights into potential therapeutic targets for managing diabetes.

Target data

Lipid-binding protein that transports phosphatidylinositol, the precursor of phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2), from its site of synthesis in the endoplasmic reticulum to the cell membrane (PubMed : 28209843). It thereby maintains the pool of cell membrane phosphoinositides, which are degraded during phospholipase C (PLC) signaling (PubMed : 28209843). Plays a key role in the coordination of Ca(2+) and phosphoinositide signaling : localizes to sites of contact between the endoplasmic reticulum and the cell membrane, where it tethers the two bilayers (PubMed : 28209843). In response to elevation of cytosolic Ca(2+), it is phosphorylated at its C-terminus and dissociates from the cell membrane, abolishing phosphatidylinositol transport to the cell membrane (PubMed : 28209843). Positively regulates insulin secretion in response to glucose : phosphatidylinositol transfer to the cell membrane allows replenishment of PI(4,5)P2 pools and calcium channel opening, priming a new population of insulin granules (PubMed : 28209843).
