Accessory component of a P4-ATPase flippase complex which catalyzes the hydrolysis of ATP coupled to the transport of aminophospholipids from the outer to the inner leaflet of various membranes and ensures the maintenance of asymmetric distribution of phospholipids. Phospholipid translocation seems also to be implicated in vesicle formation and in uptake of lipid signaling molecules. The beta subunit may assist in binding of the phospholipid substrate (Probable). Can mediate the export of alpha subunits ATP8A1, ATP8B1, ATP8B2 and ATP8B4 from the ER to the plasma membrane.

TMEM30B also known as CDC50B is a regulatory subunit of P4-ATPase flippases. This protein interacts with P4-ATPases to facilitate the transport of phospholipids from the outer to the inner leaflet of the plasma membrane. TMEM30B has a molecular mass of approximately 38 kDa. It expresses highly in brain tissue with some expression noted in the heart and skeletal muscles as well. Understanding its mechanical role helps us appreciate its importance in maintaining proper cell membrane function and integrity.
TMEM30B serves as an essential component of the P4-ATPase complex playing a role in maintaining lipid asymmetry in cell membranes. This lipid asymmetry is critical for various cellular processes including vesicular trafficking and cell signaling. TMEM30B is involved in cellular processes such as neuronal development and synaptic function given its higher expression in brain tissue. The existence of TMEM30B within this complex helps maintain the delicate balance required for functional and structural stability of cell membranes.
TMEM30B is involved in lipid metabolism and cellular homeostasis pathways. The protein's interaction with P4-ATPases like ATP8A2 plays a role in the maintenance of phospholipid distribution across the plasma membrane an important aspect of lipid metabolism. TMEM30B is also associated with the endocytosis pathway influencing how cells internalize substances and respond to external signals. It functions alongside proteins such as CDC50A in these pathways to ensure efficient cellular operations.
TMEM30B shows a relationship with neurological disorders including neurodevelopmental disorders and epilepsy. Abnormal expression or mutations in TMEM30B disrupt lipid asymmetry in neuronal cells impacting membrane fluidity and signaling leading to disease symptoms. In addition TMEM30B associates with Alzheimer's disease where it affects the amyloid precursor protein (APP) processing potentially contributing to plaque formation. Understanding these connections can lead to better therapeutic strategies targeting TMEM30B-related pathways and diseases.
