TMEM49 also known as VMP1 or Vacuole Membrane Protein 1 is an integral membrane protein with a molecular mass of approximately 52 kDa. It is located primarily within the endoplasmic reticulum and the membranes of various organelles. TMEM49/VMP1 exhibits wide expression across various tissues including the pancreas heart and skeletal muscles indicating a broad functional role in cellular processes linked to these tissues.
Biological function summary

The TMEM49/VMP1 protein plays a significant role in autophagy a cellular degradation process important for maintaining cell health. This protein is part of a complex that interacts with Beclin-1 an important player in the initiation of autophagy. By promoting the formation of autophagosomes TMEM49/VMP1 ensures the efficient degradation of unnecessary or damaged cellular components therefore facilitating cellular homeostasis and energy balance.

Pathways

TMEM49/VMP1 is important for proper functionality in autophagic and pancreatic secretory pathways. In the context of autophagy VMP1 collaborates with proteins such as LC3 and ATG proteins to progress the autophagic process. Meanwhile in pancreatic secretory pathways TMEM49/VMP1 interacts with the early secretory pathway influencing exocytosis. These pathways interact and share components highlighting the role VMP1 plays in essential cellular functions.

TMEM49/VMP1 has connections with pancreatitis and certain types of cancer. Mutations or dysregulation in VMP1 expression can contribute to the development of pancreatitis potentially through disturbances in autophagy and exocrine functions. In cancer abnormal autophagy regulation sometimes involving the VMP1-Beclin-1 complex may facilitate cancer cell survival.
