TMEM59L also known as Transmembrane Protein 59-Like is a protein with an approximate mass of 43 kDa. This protein is usually expressed in the brain with higher levels detected in regions like the cerebral cortex and hippocampus. TMEM59L is a type I transmembrane protein which means it spans the cellular membrane once and its N-terminal is inside the extracellular space. Its structure suggests a role in cellular signaling and membrane dynamics although its distinct functional domains are not entirely mapped yet.
Biological function summary

TMEM59L acts in neural differentiation and synaptic function. The protein influences neuronal development and may also play a role in synapse formation and maintenance. It is not known to be part of a larger multiprotein complex strictly but it interacts with several cellular components involved in neuronal plasticity. The presence of TMEM59L in neuronal tissue suggests that it contributes significantly to processes integral to cognitive functions.

Pathways

TMEM59L participates in cerebral processes involving cellular communication and neural integrity. It interacts within the Wnt signaling pathway which is important for neurite outgrowth and synaptic plasticity. This pathway involves other proteins such as Disheveled (DVL) and beta-catenin which further highlight the role of TMEM59L in neural connectivity and growth. In addition TMEM59L's interactions may influence pathways related to receptor trafficking connecting it to cytoskeleton-associated proteins.

TMEM59L links to neurodegenerative conditions including Alzheimer's disease and cognitive decline. In Alzheimer's disease irregularities in the expression or function of TMEM59L might disturb normal synaptic activity or neuronal health. This protein relates to beta-amyloid precursor protein (APP) which plays a role in disease pathology. Changes in TMEM59L could also affect neural plasticity leading to cognitive impairments suggesting that it might serve as a biomarker or therapeutic target in addressing such disorders.
