This data was developed using ab324735, the same antibody clone in a different buffer formulation.
Blocking and diluting buffer and concentration : 5% NFDM/TBST.
Low expression : small intestine (PMID : 10101285).
Lanes 1-2 are applied with Goat Anti-Rabbit IgG (HRP) with minimal cross reactivity with human IgG at 1/2000 and lane 3 is applied with Goat Anti-Rabbit IgG, (H+L), Peroxidase conjugated (ab97051) at 1/20000.
In Western blot, Anti-GAPDH antibody [EPR16891] - Loading Control (ab181602) staining at 1/200000 dilution.

Blocking and diluting buffer and concentration : 5% NFDM/TBST.
Performed under reducing conditions.
In Western blot, ab324735 was shown to bind specifically to TOMM34. Target of interest was observed at 34 kDa in wild-type HeLa cell lysates (lane 1) with no signal observed at this size in TOMM34 knockout cell line (lane 2) (lane 2, knockout cell line ab265546 / knockout cell lysate ab258730).
The identity of the higher MW band at approximately 50 kDa (in lane 2) is unknown.
In Western blot, Anti-GAPDH antibody [EPR16891] - Loading Control (ab181602) staining at 1/200000 dilution.
Exposure time : Lanes 1-2 : 59 seconds; Lanes 3-4 : 26 seconds.
