Flow Cytometry - Anti-TRAP/CD40L antibody [EPR20727-307] - BSA and Azide free (AB273509)
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab273499).
Flow cytometry staining of human peripheral blood mononuclear cells (PBMCs) (top) or PBMCs treated with 50ng/mL PMA and 500ng/mL Ionomycin for 18 hours (bottom), with ab273499 (right) or Recombinant Rabbit IgG, monoclonal [EPR25A] - Isotype Control (left). PBMCs were incubated for 30 mins on ice in 1x PBS containing 10 µg/ml human IgG and 10 % normal goat serum to block FC receptors and non-specific protein-protein interaction followed by the antibody ab273499 or Recombinant Rabbit IgG, monoclonal [EPR25A] - Isotype Control (1x 106 in 100 µl at 0.2 μg/ml (1/10,000 dilution)) for 30 mins on ice. The cells were simultaneously stained with CD4.
The secondary antibody Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed was incubated at 1/2000 dilution for 30 mins on ice
Acquisition of >30000 events were collected using a 50 mW Blue laser (488nm) and 525/40 bandpass filter. Events were gated on CD4+.

Flow cytometric analysis of human peripheral blood mononuclear cell (PBMC) treated with 50ng/mL PMA and 500ng/mL Ionomycin for 6h cells labelling TRAP/CD40L with ab273499 at 1/600 dilution (0.1μg) (Right panel) compared with a Rabbit monoclonal IgG (ab172730) isotype control (Left panel). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) at 1/2000 dilution was used as the secondary antibody.
Gated on viable cells.

Biological function summary
The interaction between CD40L and CD40 triggers cell proliferation differentiation and apoptosis in immune cells. CD40L contributes to the formation of an immunological synapse encompassing its role in T helper cell function. Furthermore CD40L is implicated in the activation of B cells which are necessary for antibody class switching and memory cell generation. Its broad influence shows CD40L's involvement in immune system regulation and response.
Pathways
CD40L is integral to the CD40 signaling pathway which is involved in immune and inflammatory responses. CD40L activates the NF-kB and JNK signaling pathways by engaging with CD40 which then drives the transcription of genes related to immune response. This signaling cascade impacts several other proteins such as TRAF proteins which act as intermediaries in propagating the signal.
