TRAP100 also known as MED24 is a subunit of the Mediator complex with an approximate mass of 100 kDa. It plays a significant role in transcription regulation by serving as a bridge between transcription factors and RNA polymerase II. TRAP100/MED24 is ubiquitously expressed in various tissues reflecting its central role in gene expression. It interacts with several other subunits within the Mediator complex to facilitate RNA transcription initiation.
The TRAP100/MED24 protein engages in cellular processes by working as part of the Mediator complex. This complex is essential in bridging enhancer-bound transcription factors with the basal transcriptional machinery. It plays a significant role in regulating gene expression tied to growth and development. The Mediator complex consisting of over 30 subunits including TRAP100/MED24 acts as a coactivator impacting the transcription of messenger RNA in eukaryotic cells.
This protein applies its function in transcription regulation and is essential in both Wnt and Hedgehog signaling pathways. It aids the recruitment of RNA polymerase II which is important for the initiation phase in these pathways. Interactions with proteins such as β-catenin and Gli proteins reveal the involvement of TRAP100/MED24 in transcriptional regulation downstream of these pathways influencing gene networks involved in cell fate determination and tissue homeostasis.
Defective TRAP100/MED24 function links to conditions such as cancer and neurodevelopmental disorders.
