Activity summary

TRAPPC2 also known as sedlin is a component of the transport protein particle (TRAPP) complex with a molecular mass of approximately 17 kDa. It is a small protein that has a significant role in vesicular trafficking within cells. TRAPPC2 is expressed widely in human tissues with notable levels in the testis skeletal muscle and heart. This protein participates in the regulation of intracellular transport processes by assembling into multi-subunit complexes that facilitate the movement of vesicles.

Biological function summary

TRAPPC2 plays a role in the formation and function of the TRAPP complex which is fundamental for intracellular vesicle trafficking. The TRAPP complex exists in different forms and participates in the tethering of vesicles to target membranes. TRAPPC2’s involvement in this complex influences ER-Golgi transportation impacting how proteins and lipids move inside cells. By working as part of the TRAPP complex TRAPPC2 contributes to the maintenance of cellular homeostasis by controlling the flow and direction of cargo.

Pathways

TRAPPC2 is involved in the secretory pathway which is essential for protein sorting and transport. It interacts with other TRAPP subunits such as TRAPPC3 and TRAPPC9 to ensure effective vesicle fusion and trafficking processes. In addition to the secretory pathway TRAPPC2 is engaged in the autophagy pathway. Through its participation in these pathways TRAPPC2 affects how cells manage and recycle cellular components under varying conditions.

Associated diseases and disorders

Mutations in TRAPPC2 are linked to X-linked spondyloepiphyseal dysplasia tarda a skeletal disorder affecting bone growth and development. The deficiency of TRAPPC2 disrupts normal vesicular trafficking ultimately leading to the symptoms observed in this disorder. TRAPPC2’s interactions with proteins such as TRAPPC3 and TRAPPC9 through these pathways underline its significance in maintaining cellular functions that when altered result in specific pathological states.
