TRAPPC6B also known as Trafficking Protein Particle Complex Subunit 6B is a component of the TRAPP complex which plays a role in vesicle-mediated transport within cells. TRAPPC6B is a protein with a molecular mass of approximately 16 kDa. It is expressed in various tissues including the liver kidney and brain indicating its broad involvement in cellular processes across different cell types.
Biological function summary

TRAPPC6B is involved in regulating intracellular trafficking particularly in the early steps of the secretory pathway. As part of the TRAPP complex it contributes to the tethering of COPII vesicles to Golgi membranes facilitating transport between the endoplasmic reticulum and Golgi apparatus. This action helps maintain cellular homeostasis and proper distribution of proteins and lipids within cells.

Pathways

TRAPPC6B is significant in the ER-Golgi transport pathway where it interacts with other subunits of the TRAPP complex including TRAPPC2 and TRAPPC3. These interactions are important for the assembly and function of the TRAPP complex in vesicle tethering. TRAPPC6B also plays a role in the macroautophagy pathway affecting the biogenesis of autophagosomes which are essential for cellular degradation and recycling processes.

TRAPPC6B has been implicated in neurodevelopmental disorders and is linked to intellectual disability. Mutations in TRAPPC6B can disrupt vesicle transport mechanisms leading to developmental delays and neurological symptoms. TRAPPC6B interacts with various proteins involved in these conditions like Sec23 and Sec24 which are subunits of the COPII coat. These interactions highlight the importance of TRAPPC6B in neurodevelopmental health and provide insights into possible therapeutic targets for related disorders.

Target data

Component of a transport protein particle (TRAPP) complex that may function in specific stages of inter-organelle traffic (PubMed : 16025134, PubMed : 16828797). Specifically involved in the early development of neural circuitry, likely by controlling the frequency and amplitude of intracellular calcium transients implicated in the regulation of neuron differentiation and survival (Probable).
