TRIB1 also known as tribbles homolog 1 is a pseudo kinase protein with a mass of approximately 41 kDa. It lacks catalytic activity typically expected from kinases but it plays a critical mechanical role via its protein interactions. TRIB1 is expressed in various human tissues including liver spleen and heart. It acts mainly through modulating protein degradation and transcription regulation processes.
TRIB1 impacts cell signaling and metabolic processes by forming complexes with other proteins. It exerts a regulatory role by binding to E3 ubiquitin ligases influencing the degradation of target proteins. TRIB1 also interacts with transcription factors affecting gene expression. These actions highlight its involvement in maintaining cellular homeostasis by controlling inflammatory responses and metabolic pathways.
TRIB1 influences metabolic and signaling pathways by interacting with key regulatory proteins. It plays a significant role within the MAP kinase pathway and the PI3K-AKT pathway. Both pathways are important for cell proliferation survival and metabolism. In these pathways proteins like ATF4 in the MAP kinase pathway and AKT1 in the PI3K-AKT pathway associate with TRIB1 allowing it to exert control over important cellular processes.
TRIB1 is linked to conditions such as cardiovascular disease and certain cancers. Its regulatory function within metabolic pathways connects it to lipid metabolism disorders impacting cardiovascular health. TRIB1 also relates to leukemia through its interactions with the transcription factor C/EBPα. Dysregulation of TRIB1 expression or function can disrupt normal cellular functions leading to disease progression.
