Supplementary information

TRIM3 also known as Tripartite Motif Containing 3 is a member of the tripartite motif family of proteins and has a mass of approximately 70 kDa. This protein functions mechanically as an E3 ubiquitin-protein ligase involved in the tagging of substrates for proteasomal degradation. TRIM3 is expressed in various tissues though it shows higher levels in the brain particularly in neurons. The presence of TRIM3 in neuronal tissues suggests an important role in neural function and homeostasis.
Biological function summary

TRIM3 influences key cellular processes such as cell cycle progression and apoptosis. It is part of a multimeric protein complex that regulates the degradation of certain protein substrates. By modulating the ubiquitin-proteasome system TRIM3 contributes to maintaining protein homeostasis within cells. The ubiquitin ligase activity of TRIM3 ensures that misfolded or damaged proteins are correctly identified and directed towards degradation which is essential for normal cell function.

Pathways

TRIM3 integrates into critical signaling networks that maintain cellular balance and response. It plays a significant role in the ubiquitin-proteasome pathway which is a major cellular mechanism for controlling protein levels. TRIM3 interacts with several proteins in this pathway including the ubiquitin-conjugating enzymes to exert its regulatory effects. Additionally TRIM3 has connections with other pathways involved in neural development and function intersecting with proteins such as neurofilament chains.

TRIM3 has been linked to neurodegenerative conditions and cancer. In Alzheimer's disease for example TRIM3 associates with tau protein possibly affecting its degradation and accumulation which contributes to disease pathology. There is also evidence of TRIM3 involvement in glioblastoma where its regulatory role in protein turnover may influence cell proliferation and tumor progression. The modulation of TRIM3 activity or its interactions in these conditions could provide valuable insights into therapeutic targets.

Target data

E3 ubiquitin ligase that plays essential roles in neuronal functions such as regulation of neuronal plasticity, learning, and memory (By similarity). In addition to its neuronal functions, participates in other biological processes such as innate immunity or cell cycle regulation. Component of the cytoskeleton-associated recycling or transport complex in neurons, polyubiquitinates gamma-actin, thus regulating neuronal plasticity, learning, and memory (By similarity). Ubiquitinates postsynaptic scaffold GKAP, a neuronal substrate involved in synaptic remodeling and thereby modulates dendritic spine morphology (By similarity). Positively regulates motility of microtubule-dependent motor protein KIF21B (By similarity). Induces growth arrest via its RING-dependent E3 ligase activity and ubiquinates CDKN1A (PubMed : 24393003). Positively regulates TLR3-mediated signaling by mediating 'Lys-63'-linked polyubiquitination of TLR3 (PubMed : 32878999). In turn, promotes the recognition and sorting of polyubiquitinated TLR3 by the ESCRT complexes (PubMed : 32878999).
