TRIM30 also known as Rpt1 is a member of the tripartite motif (TRIM) family of proteins characterized by the presence of a RING finger B-box and coiled-coil domains. In humans TRIM30 has a molecular mass of about 56 kDa. The protein primarily resides in the cytoplasm and nucleus suggesting its involvement in various cellular processes. Expression of TRIM30 is observed across different tissues with varying levels depending on cell type and tissue-specific functions.
Biological function summary

The protein TRIM30 acts as an E3 ubiquitin ligase an important component in the regulation of protein degradation processes. It facilitates the attachment of ubiquitin molecules to specific substrate proteins targeting them for proteasomal degradation and modulating various cellular activities. TRIM30 associates with several complexes involved in immune responses and acts as a regulatory factor in innate immunity. Through ubiquitination TRIM30 influences signaling pathways and cellular responses to external stimuli maintaining cellular homeostasis.

Pathways

TRIM30 influences signal transduction by interacting with important molecules in pathways like the NF-kB signaling and MAPK pathways. In the NF-kB signaling pathway TRIM30 can modulate the activation of NF-kB impacting the expression of pro-inflammatory cytokines and immune responses. Within the MAPK pathway this modulation affects cellular responses like proliferation and apoptosis. TRIM30 thereby interacts with other proteins such as TAB2 and TAK1 shaping the downstream consequences of these pathways.

TRIM30 has connections with conditions related to immune dysregulation and inflammation such as systemic lupus erythematosus and rheumatoid arthritis. Its role in controlling inflammatory signaling makes it significant in the progression of these disorders. Within lupus for example abnormal regulation of TRIM30 might contribute to hyperactive immune responses. TRIM30's involvement in these diseases links it to proteins such as NF-kB and other components in inflammatory pathways dictating the severity and progression of the conditions.

Target data

Trans-acting factor that regulates gene expression of interleukin 2 receptor alpha chain. May affect IL2R-alpha expression through cis-acting negative regulatory elements or through competition with proteins that bind to enhancer or activator sequences. Negatively regulates Toll-like receptor (TLR)-mediated activation of NFKB by promoting degradation of TAB2 and TAB3 and preventing TRAF6 autoubiquitination. Negatively regulates production of reactive oxygen species (ROS) which inhibits activation of the NLRP3 inflammasome complex. This, in turn, regulates activation of CASP1 and subsequent cleavage of IL1B and IL18. No activity detected against a range of retroviruses including a number of lentiviruses, gammaretroviruses and betaretroviruses.
