TRIM43 also known as Tripartite Motif Containing 43 is a protein with a molecular weight of approximately 56 kDa. This protein belongs to the tripartite motif (TRIM) family known for containing a RING finger B-box type 1 and 2 and a coiled-coil domain. TRIM43 plays a role in ubiquitination processes and is highly expressed in the testes. Several studies have reported its presence at lower levels in other tissues suggesting a more specialized function in reproductive biology.
Biological function summary

Functions involving TRIM43 include roles in modulating innate immune responses. While not part of a large multi-protein complex TRIM43 interacts closely with specific cellular pathways involved in immune regulation. It participates in the restriction of viral activity by targeting viral proteins for degradation therefore influencing viral replication processes. This highlights its critical function in cellular defense mechanisms.

Pathways

The ubiquitination regulation carried out by TRIM43 integrates into innate immune signaling pathways. TRIM43 interacts with pathways such as the toll-like receptor (TLR) signaling pathway which is essential for innate immune responses. It also relates to the regulation of the NF-kB pathway an important factor for immune response and inflammation. TRIM43's functional dynamics may interact with other TRIM proteins contributing to the broader modulation of these pathways.

Expanded investigations have connected TRIM43 dysregulation with cancer specifically germ cell tumors. The protein's function in immune modulation suggests potential involvement in autoimmunity like systemic lupus erythematosus. Changes in TRIM43 expression levels especially in testicular tissues associate with certain cancerous transformations. Interactions with proteins like IFI16 and DDX3X may also amplify these disease links reflecting its impact in disease mechanisms and potential as a target for therapeutic solutions.

Product protocols

For this product, it's our understanding that no specific protocols are required. You can visit:

Target data

E3 ligase that regulates nuclear lamina integrity and the association of viral chromatin with transcriptionally-active host chromatin. Acts thereby as a herpesvirus-specific antiviral factor and mediates the ubiquitination-dependent proteasomal degradation of PCNT.
