TRIM44 also known as RNF104 is a member of the tripartite motif (TRIM) family of proteins. It has a molecular mass of approximately 67 kDa. TRIM44 is found in various tissues including the lungs liver kidney and muscle. Its mechanical role involves functioning as an E3 ubiquitin ligase facilitating the transfer of ubiquitin to specific protein substrates which tags them for degradation or alters their function and location. The C-terminal RING domain of TRIM44 is important for its ubiquitin ligase activity.
Biological function summary

TRIM44 participates in cell proliferation migration and differentiation. It plays a role in maintaining cellular homeostasis by regulating protein turnover and stability. TRIM44 does not form part of a larger complex but operates independently. By ensuring proper degradation and recycling of proteins it impacts various cellular processes and is linked to cellular stress responses. Its regulatory functions extend to influence various biological processes critical for normal cellular operation.

Pathways

TRIM44 interacts with the ubiquitin-proteasome system and the PI3K/AKT signaling pathway. In the ubiquitin-proteasome system TRIM44 ubiquitinates substrates for proteasomal degradation ensuring cellular protein quality control. Through the PI3K/AKT pathway TRIM44 influences cell survival and growth by modulating signaling proteins within the pathway such as AKT and mTOR. TRIM44's activity within these pathways highlights its role in cellular homeostasis and response to growth signals.

TRIM44 has been implicated in cancer and neurodegenerative diseases. In various cancers such as ovarian and hepatocellular carcinoma TRIM44 overexpression has linked to increased cancer cell proliferation and invasion often through its modulation of the PI3K/AKT pathway. Additionally in neurodegenerative diseases TRIM44 influences protein quality control connecting to proteins involved in proteostasis such as Parkin in Parkinson’s disease. These associations suggest TRIM44's activity correlates with disease progression marking it as a potential target for therapeutic intervention.
