TRIM45 also known as tripartite motif-containing protein 45 belongs to the TRIM/RBCC protein family. It acts mechanically as an E3 ubiquitin-protein ligase facilitating the attachment of ubiquitin to substrate proteins a process vital for proteasomal degradation. TRIM45 has an approximate mass of 55 kDa. Expression of TRIM45 occurs in various tissues particularly in the brain. It contains a RING finger domain one B-box type zinc finger and a coiled-coil region defining its structure.
Biological function summary

TRIM45 functions in the regulation of transcriptional activity and cell signaling pathways. It operates independently and may also form part of a larger protein complex during these activities. TRIM45 plays a role in cellular stress responses and modulates transcription factors through its ubiquitinligase activity thereby influencing gene expression. By controlling these processes TRIM45 impacts cellular proliferation and survival.

Pathways

TRIM45 participates in critical signaling pathways such as the MAPK/ERK pathway and the Wnt signaling pathway. In the MAPK/ERK pathway TRIM45 interacts with various kinases to regulate cell growth and differentiation. In the Wnt signaling pathway TRIM45 associates with proteins like β-catenin to influence cell fate decisions. These connections enable TRIM45 to coordinate cellular responses to environmental stimuli impacting many cellular processes.

TRIM45 has a connection to neurodegenerative diseases and certain cancers. Alterations in TRIM45 expression or function may contribute to the pathogenesis of diseases like glioblastoma.
