Supplementary information

TRIM67 also known as Tripartite Motif Containing 67 is a protein encoded by the TRIM67 gene. It belongs to the tripartite motif (TRIM) family of proteins characterized by an N-terminal RING finger domain one or two B-box motifs and a coiled-coil region. TRIM67 has a molecular mass of approximately 53 kDa. This protein is mainly expressed in the brain particularly in neurons highlighting its importance in neural tissues.
Biological function summary

TRIM67 plays an important role in neuronal development and differentiation. It contributes to processes like axon outgrowth and guidance which are critical for proper neuronal network formation. TRIM67 does not function as part of a complex but interacts with other proteins for executing its roles. It is known to modulate several signaling pathways key to cell proliferation and survival emphasizing its importance in cellular maintenance and integrity.

Pathways

TRIM67 impacts the regulation of the Wnt signaling pathway which is essential for numerous developmental processes. Moreover it has a relationship with the PI3K/AKT signaling pathway which is important for cell growth and apoptosis. In these pathways TRIM67 can interact with proteins like DVL and AKT1 contributing to their regulation and overall pathway stability. This connectivity underlines the multifaceted role TRIM67 plays in cell signaling and developmental biology.

TRIM67 has been associated with neurological disorders given its strong expression in neuronal tissues. Specifically it relates to conditions like autism spectrum disorders (ASD) where disruptions in its expression or mutations could impact neural connectivity. Additionally neurodegenerative diseases such as Alzheimer's disease may also involve TRIM67 due to its role in neuronal survival and function. These associations highlight a connection with proteins like APP and BDNF which are linked to neural health and disease.
