Function

Relays signals from the cell surface to the nucleus to weaken adherens junction and promote actin cytoskeleton reorganization and cell invasiveness. Involved in lysophosphatidic acid-induced cell adhesion and migration. Acts as a transcriptional coactivator for NF-kappa-B and JUN, and mediates the transrepression of these transcription factors induced by glucocorticoid receptor.

Activity summary

TRIP6 also known as Thyroid Hormone Receptor Interactor 6 or ZRP-1 (Zyxin Related Protein 1) is a cytoskeletal adaptor protein with a molecular mass of approximately 55 kDa. Its primary function involves acting as a scaffold protein that facilitates interactions between several intracellular signaling molecules. TRIP6 localizes mainly to focal adhesion sites and the actin cytoskeleton where it regulates cell motility and organization. This protein is usually expressed in various tissues but expression levels are notably prominent in the lymphoid tissues and the gastrointestinal tract.

Biological function summary

The mechanical interactions of TRIP6 influence cell migration adhesion and morphogenesis. It functions as a component of the focal adhesion complex where it plays an important role in linking the actin cytoskeleton to integrin receptors. Additionally TRIP6 participates in the regulation of gene expression by translocating to the nucleus and interacting with transcription factors to modulate specific gene targets.

Pathways

The involvement of TRIP6 is significant within signaling pathways such as the RhoA pathway and the NF-kB signaling pathway. Through RhoA TRIP6 contributes to the modulation of cell shape and motility impacting cellular processes such as angiogenesis and wound healing. In the NF-kB pathway TRIP6 can influence inflammatory responses by affecting gene transcription. TRIP6 is associated with proteins like paxillin and focal adhesion kinase (FAK) which are key components in these pathways.

Associated diseases and disorders

TRIP6 has notable implications in cancer progression and inflammatory diseases. It has been implicated as a contributor to tumorigenesis due to its role in cell motility and invasion often showing altered expression in various cancers such as breast and prostate cancer. TRIP6 also interacts with the protein zyxin and this interaction influences cancer cell metastasis. Additionally TRIP6 can affect inflammatory responses in disorders such as inflammatory bowel disease where its modulation impacts cellular pathways related to inflammation and immune response.
