TrkA TrkB and TrkC also known as Ntrk1 Ntrk2 and Ntrk3 respectively are receptor tyrosine kinases expressed mainly in neurons of the central and peripheral nervous systems. TrkA has a mass of about 140 kDa TrkB about 145 kDa and TrkC around 140 kDa. These receptors bind to neurotrophins: TrkA interacts with nerve growth factor (NGF) TrkB with brain-derived neurotrophic factor (BDNF) and neurotrophin-4 (NT-4) and TrkC with neurotrophin-3 (NT-3). Their primary role is to mediate neurotrophin signaling resulting in activation of downstream signaling cascades that support neuronal survival development and function.
Biological function summary

TrkA TrkB and TrkC play critical roles in nervous system development and maintenance. These receptors upon binding with their respective ligands form dimers which leads to autophosphorylation on specific tyrosine residues. This activation recruits adaptor proteins and through a series of interactions initiates multiple signaling pathways. Among these are the MAPK PI3K and PLCγ pathways which further propagate signals that regulate cell differentiation growth and survival demonstrating their importance in neurobiology.

Pathways

These Trk receptors engage in important signaling pathways that include the MAPK/ERK pathway and the PI3K/Akt pathway. These pathways are major components of cellular functions like proliferation differentiation and survival. During activation Trk receptors can interact with proteins like SHC and GRB2 within these pathways. Their signaling cascades influence neuronal plasticity and repair indicating their vital involvement in cellular responses to environmental stimuli.

TrkA TrkB and TrkC serve important roles in conditions like neurodegenerative diseases and certain cancers. For instance altered Trk receptor signaling has been associated with Alzheimer's disease where BDNF signaling via TrkB is often impaired. Similarly mutations or dysfunctional signaling of these receptors are implicated in cancers such as neuroblastoma where TrkA can act as an oncogene. These disorders highlight the relationship of these receptors to other proteins like tau in Alzheimer's disease and p75NTR in neuronal growth and cancerous transformations.

Receptor tyrosine kinase involved in the development and the maturation of the central and peripheral nervous systems through regulation of proliferation, differentiation and survival of sympathetic and nervous neurons. High affinity receptor for NGF which is its primary ligand (PubMed : 1281417, PubMed : 15488758, PubMed : 17196528, PubMed : 1849459, PubMed : 1850821, PubMed : 22649032, PubMed : 27445338, PubMed : 8325889). Can also bind and be activated by NTF3/neurotrophin-3. However, NTF3 only supports axonal extension through NTRK1 but has no effect on neuron survival (By similarity). Upon dimeric NGF ligand-binding, undergoes homodimerization, autophosphorylation and activation (PubMed : 1281417). Recruits, phosphorylates and/or activates several downstream effectors including SHC1, FRS2, SH2B1, SH2B2 and PLCG1 that regulate distinct overlapping signaling cascades driving cell survival and differentiation. Through SHC1 and FRS2 activates a GRB2-Ras-MAPK cascade that regulates cell differentiation and survival. Through PLCG1 controls NF-Kappa-B activation and the transcription of genes involved in cell survival. Through SHC1 and SH2B1 controls a Ras-PI3 kinase-AKT1 signaling cascade that is also regulating survival. In absence of ligand and activation, may promote cell death, making the survival of neurons dependent on trophic factors.. Isoform TrkA-III. Resistant to NGF, it constitutively activates AKT1 and NF-kappa-B and is unable to activate the Ras-MAPK signaling cascade. Antagonizes the anti-proliferative NGF-NTRK1 signaling that promotes neuronal precursors differentiation. Isoform TrkA-III promotes angiogenesis and has oncogenic activity when overexpressed.
