TRPML3 also known as Mucolipin-3 or MCOLN3 is a member of the transient receptor potential (TRP) channel family. It functions as a non-selective cation channel that plays an important role in calcium ion homeostasis within cells. The TRPML3 protein has a molecular mass of approximately 63 kDa. It is expressed in various tissues mainly in sensory hair cells of the inner ear as well as in other tissues like the retina and the kidneys. Its distribution suggests its involvement in specific cellular processes and functions.
Biological function summary

TRPML3 plays an important role in the maintenance of lysosomal ion homeostasis and cellular signaling. It is an integral part of lysosomal membranes and contributes to the regulation of lysosomal trafficking. This channel is involved in the regulation of intracellular calcium levels and phosphoinositide metabolism. TRPML3 functions individually but may also interact with other members of the TRP channel family contributing to its functional and biological diversity.

Pathways

TRPML3 is involved in the lysosome-related pathways which include autophagy and endo-lysosomal trafficking. These processes are critical for cellular homeostasis recycling of cellular materials and response to stress. TRPML3 shows a functional connection with other TRP channels such as TRPML1 and TRPML2 in these pathways. It is also associated with the phosphoinositide metabolic pathway influencing diverse cellular activities such as membrane trafficking and signal transduction.

Mutations in TRPML3 have been linked to hearing impairment and vision disorders such as deafness and Agammaglobulinemia. These conditions are often ascribed to the malfunctioning of the channel leading to disrupted ion-homeostasis and cellular signaling. Research has also shown that TRPML3 may interact with proteins involved in these disorders including its close relatives TRPML1 and TRPML2 which have been studied for their role in similar lysosomal storage disorders.

Nonselective cation channel probably playing a role in the regulation of membrane trafficking events. Acts as a Ca(2+)-permeable cation channel with inwardly rectifying activity (PubMed : 18369318, PubMed : 19497048, PubMed : 19522758, PubMed : 19885840, PubMed : 29106414). Mediates release of Ca(2+) from endosomes to the cytoplasm, contributes to endosomal acidification and is involved in the regulation of membrane trafficking and fusion in the endosomal pathway (PubMed : 21245134). Also permeable to Mg(2+), Na(+) and K(+) (By similarity). Does not seem to act as mechanosensory transduction channel in inner ear sensory hair cells. Proposed to play a critical role at the cochlear stereocilia ankle-link region during hair-bundle growth (By similarity). Involved in the regulation of autophagy (PubMed : 19522758). Through association with GABARAPL2 may be involved in autophagosome formation possibly providing Ca(2+) for the fusion process (By similarity). Through a possible and probably tissue-specific heteromerization with MCOLN1 may be at least in part involved in many lysosome-dependent cellular events (PubMed : 19885840). Possible heteromeric ion channel assemblies with TRPV5 show pharmacological similarity with TRPML3 (PubMed : 23469151).
