Immunohistochemical analysis of paraffin-embedded human dorsal root ganglia tissue labeling TRPV1 with ab305178 at 1/100 dilution (9.85 µg/ml) followed by ready to use LeicaDS9800 (Bond® Polymer Refine Detection). Positive staining on human dorsal root ganglia. The section was incubated with ab305178 for 30 mins at room temperature, followed by anti-mouse IgG1 antibody (ab125913) for 8 mins during the LeicaDS9800 kit staining procedure. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin. Secondary antibody only control : Secondary antibody is ready to use LeicaDS9800 (Bond® Polymer Refine Detection). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution2) for 20 mins.

Immunohistochemical analysis of 4% PFA-fixed, 0.2% Triton X-100 permeabilized frozen rat DRG (fresh) tissue labeling TRPV1 with ab305178 at 1/50 dilution followed by ab150117 Goat Anti-Mouse IgG H&L (Alexa Fluor® 488) preadsorbed at 1/1000 dilution (2 µg/mL) (Green). Confocal image showing positive staining on rat DRG. The section was incubated with ab305178 for 60 mins at room temperature. The section was then mounted using Fluoromount®. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8). The nuclear counterstain was DAPI (Blue). The receptor operates as part of a homotetrameric complex contributing to neural signaling. It is activated by chemical ligands such as capsaicin the compound responsible for the spicy sensation in chili peppers influencing pain and inflammation pathways.
Pathways
TRPV1 plays essential roles in pain and nociception pathways. It intersects with the inflammatory pathway where its activity is modulated by protein kinase C (PKC) and phospholipase C (PLC). These pathways involve calcium signaling and relate to proteins such as PKA and calcineurin which modulate TRPV1 activity through phosphorylation and dephosphorylation affecting the body's response to noxious stimuli.
