Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-TRPV5 antibody [EPR8875] (AB137028)
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human kidney tissue sections labeling TRPV5 with purified ab137028 at 1 : 300 dilution (0.95 µg/ml). Heat mediated antigen retrieval was performed using Bond ™ Epitope Retrieval Solution 1 (pH 6.0). Rabbit specific IHC polymer detection kit HRP/DAB (ab209101) was used as the secondary antibody. Negative control : PBS instead of the primary antibody. Hematoxylin was used as a counterstain.

We have systematically measured KD (the equilibrium dissociation constant between the antibody and its antigen), of more than 840 recombinant antibodies to assess not only their individual KD values but also to see the average affinity of antibody. Based on the comparison with published literature values for mouse monoclonal antibodies, Recombinant antibodies appear to be on average 1-2 order of magnitude higher affinity.

Biological function summary
TRPV5 operates as a critical component in the regulation of calcium homeostasis. It exists as a monomer but often forms functional complexes with other calcium-transporting proteins to fulfill its role. TRPV5 modulates the amount of calcium that is reabsorbed from urine back into the bloodstream a process essential for maintaining proper levels of calcium in the body and supporting various physiological functions including muscle contraction and nerve conduction.
Pathways
TRPV5 plays a significant role in the renal calcium reabsorption pathway. By facilitating the entry of calcium ions into kidney epithelial cells TRPV5 works in conjunction with calbindin-D28K a calcium-binding protein which buffers intracellular calcium levels. Another important pathway involving TRPV5 is vitamin D-mediated regulation of calcium balance where vitamin D enhances the expression of TRPV5 in response to calcium requirements in the body.
