Paraffin-embedded human placenta tissue stained for TRPV6 using ab238107 at 1/300 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum for 30 minutes at room temperature. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

TRPV6 also known as transient receptor potential cation channel subfamily V member 6 is a calcium-selective channel with a mass of approximately 89 kDa. This channel plays an important role in mediating calcium entry across epithelial tissues. TRPV6 is expressed largely in epithelial cells of organs like the intestine kidney and placenta. The channel's main function is to regulate calcium absorption by epithelial cells enabling fine control of intracellular calcium levels.
Biological function summary

TRPV6 allows the transport of calcium ions into the cells influencing cellular calcium balance. It can be considered a part of the epithelial calcium channel complex together with TRPV5. Calcium selective cation channel that mediates Ca(2+) uptake in various tissues, including the intestine (PubMed : 11097838, PubMed : 11248124, PubMed : 11278579, PubMed : 15184369, PubMed : 23612980, PubMed : 29258289). Important for normal Ca(2+) ion homeostasis in the body, including bone and skin (By similarity). The channel is activated by low internal calcium level, probably including intracellular calcium store depletion, and the current exhibits an inward rectification (PubMed : 15184369). Inactivation includes both a rapid Ca(2+)-dependent and a slower Ca(2+)-calmodulin-dependent mechanism; the latter may be regulated by phosphorylation. In vitro, is slowly inhibited by Mg(2+) in a voltage-independent manner. Heteromeric assembly with TRPV5 seems to modify channel properties. TRPV5-TRPV6 heteromultimeric concatemers exhibit voltage-dependent gating.
