Supplementary information

TSPAN6 also known as Tetraspanin-6 is a member of the tetraspanin protein family. This protein has a molecular mass of approximately 27 kDa. TSPAN6 is an integral membrane protein and typically resides in the plasma membrane. The protein is broadly expressed across a range of tissues including brain immune cells and various epithelial tissues. Tetraspanins generally play a role in organizing the structure of the cell surface membrane and TSPAN6 is believed to contribute to this structural framework as well.
Biological function summary

TSPAN6 participates in various cell processes by influencing the molecular interactions at the cell surface. As a member of the tetraspanin family it associates with other transmembrane proteins to form complexes. These complexes can regulate cell signaling adhesion and migration. As a result TSPAN6 plays a part in modulating immune response and cellular development processes. Its involvement in the packaging of molecules like other tetraspanins is important for processes such as cell communication and motility.

Pathways

TSPAN6 takes part in several signaling pathways including the Wnt signaling pathway. It interacts with other proteins like the beta-catenin to influence cell fate decisions and proliferation. TSPAN6 may also have a role within the regulation of the immune system pathways potentially through interaction with proteins such as integrins. This involvement can affect processes like immune surveillance and response to pathogens.

Aberrant expression of TSPAN6 has connections with neurological conditions and cancers. In neurodevelopmental disorders such as autism spectrum disorders changes in TSPAN6 expression may interfere with neural connectivity and communication. This protein has been implicated in cancer progression especially in colon cancer where it might modulate cell proliferation and migration. In these conditions TSPAN6 could be intertwined with proteins like EGFR affecting cellular growth and survival.
