Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-TTC11/FIS1 antibody (AB229969)
Paraffin-embedded human kidney tissue stained for TTC11/FIS1 using ab229969 at 1/600 dilution in immunohistochemical analysis.
After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Immunocytochemistry/ Immunofluorescence - Anti-TTC11/FIS1 antibody (AB229969)
Immunofluorescent analysis of HeLa (human epithelial cell line from cervix adenocarcinoma) cells stained for TTC11/FIS1 (green) using ab229969 at 1/100 dilution in ICC/IF, followed by Alexa Fluor 488® congugated Goat Anti-Rabbit IgG (H+L).

Immunoprecipitation - Anti-TTC11/FIS1 antibody (AB229969)
TTC11/FIS1 was immunoprecipitated from 500 μg MCF7 (human breast adenocarcinoma cell line) whole cell lysate with 4 μg of ab229969.
Lane 1 : Rabbit monoclonal IgG (1 μg) IP instead of ab229969 in MCF7 whole cell lysate.
Lane 2 : ab229969 IP in MCF whole cell lysate.
Lane 3 : MCF7 whole cell lysate 20 ug (input).
For western blotting, an HRP-conjugated light chain specific antibody was used as the Secondary antibody at 1/50000 dilution.

Western blot - Anti-TTC11/FIS1 antibody (AB229969)
Western blot : Rabbit Polyclonal to TTC11/FIS1 229969 staining at 1/2000 dilution, shown in green; Mouse anti GAPDH (ab8245) loading control staining at 1/20,000 dilution, shown in magenta. A band was observed at 16 kDa in Wild-type Jurkat cell lysates with no signal observed at this size in FIS1 knockout Jurkat cell line. To generate this image, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3pc Milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4°C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit 800CW & Goat anti-Mouse 680RD at 1/20,000 dilution.

Biological function summary
FIS1 is critical in controlling mitochondrial size and quantity. It is part of a complex that includes proteins like DRP1 which is heavily involved in promoting the division of mitochondria. By working with DRP1 FIS1 helps maintain cellular energy levels and dynamics by ensuring that mitochondria are appropriately divided and distributed. This role impacts overall cell health and function.
Pathways
FIS1 is integral to the pathways regulating mitochondrial fission and fusion. It interacts closely within the pathway involving DRP1 and MFF both of which are critical in these processes. Furthermore FIS1's interactions in these pathways help balance mitochondrial fission and fusion maintaining cellular energy homeostasis and proper mitochondrial function.
