Immunocytochemistry/ Immunofluorescence - Anti-TTC11/FIS1 antibody (AB96764)
ICC/IF image of ab96764 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab96764, 1µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h.Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Biological function summary
FIS1 is critical in controlling mitochondrial size and quantity. It is part of a complex that includes proteins like DRP1 which is heavily involved in promoting the division of mitochondria. By working with DRP1 FIS1 helps maintain cellular energy levels and dynamics by ensuring that mitochondria are appropriately divided and distributed. This role impacts overall cell health and function.
Pathways
FIS1 is integral to the pathways regulating mitochondrial fission and fusion. It interacts closely within the pathway involving DRP1 and MFF both of which are critical in these processes. Furthermore FIS1's interactions in these pathways help balance mitochondrial fission and fusion maintaining cellular energy homeostasis and proper mitochondrial function.
