Required for the preservation of the structural and functional integrity of mitochondrial respiratory complex III by allowing the physiological turnover of the Rieske protein UQCRFS1 (PubMed:21278747, PubMed:28673544). Involved in the clearance of UQCRFS1 N-terminal fragments, which are produced upon incorporation of UQCRFS1 into the complex III and whose presence is detrimental for its catalytic activity (PubMed:28673544).

TTC19 also known as Tetratricopeptide Repeat Domain 19 operates mechanically as a nuclear-encoded mitochondrial protein involved in mitoribosome assembly and function. This protein has a mass of approximately 45 kDa. Researchers have identified its presence mainly in mitochondria-rich tissues such as the brain and heart. The expression pattern suggests its significant role in tissues with high energy demand.
This protein plays an important role in regulating mitochondrial respiratory chain complex III assembly. TTC19 facilitates the proper incorporation of the UQCRB subunit into complex III essential for the electron transport chain. It belongs to a multi-subunit complex within the inner mitochondrial membrane. This complex ensures optimal mitochondrial performance highlighting TTC19's role in cellular energy homeostasis.
TTC19 influences mitochondrial and oxidative phosphorylation pathways. These pathways are critical for ATP production in cell metabolism. TTC19 interacts with proteins such as UQCRC1 during the electron transport process. By enabling efficient electron flow TTC19 supports cellular respiration and energy conversion pathways essential for cell viability.
Functional deficiencies or mutations in this target link to diseases like mitochondrial complex III deficiency and Leigh Syndrome. These conditions often manifest with neurological symptoms and multisystemic involvement. Proteins such as cytochrome c can become indirectly affected by TTC19 defects leading to compromised electron transport and cellular energy production ultimately contributing to disease etiology.
