Component of a complex required to localize phosphatidylinositol 4-kinase (PI4K) to the plasma membrane. The complex acts as a regulator of phosphatidylinositol 4-phosphate (PtdIns(4)P) synthesis. In the complex, plays a central role in bridging PI4KA to EFR3B and HYCC1, via direct interactions (PubMed:26571211).

TTC7B also known as Tetratricopeptide Repeat Domain 7B is a protein with a mass of approximately 114 kDa. Mechanically it contains several tetratricopeptide repeat (TPR) motifs that facilitate protein-protein interactions. This protein is expressed in many tissues with notable levels in the liver and kidney. Its role is to help with various cellular processes likely through binding with other proteins to form functional complexes.
TTC7B contributes to cellular functions through its involvement in the phosphatidylinositol-4 kinase pathway. This pathway is important for the regulation of cell signaling and membrane trafficking. TTC7B is part of the GOLPH3/TTC7B complex which maintains proper Golgi function and morphology an essential aspect for vesicular transport and signal transduction.
The phosphatidylinositol signaling and inositol phosphate metabolism pathways require TTC7B's participation. These pathways are critical for various cellular processes including cell growth survival and communication. TTC7B interfaces with proteins such as PI4KA and FAM126A which are important for phosphatidylinositol 4-phosphate synthesis impacting overall pathway functionality.
Improper TTC7B function can associate with Golgi function-related disorders. Conditions such as Lenz-Majewski hyperostotic dwarfism have connections to the dysregulation of pathways involving TTC7B. In these contexts proteins like PI4KA also interact in the disease mechanism illustrating TTC7B's significance in maintaining cellular and physiological balance.
