Tubulin is the major constituent of microtubules, a cylinder consisting of laterally associated linear protofilaments composed of alpha- and beta-tubulin heterodimers. Microtubules grow by the addition of GTP-tubulin dimers to the microtubule end, where a stabilizing cap forms. Below the cap, tubulin dimers are in GDP-bound state, owing to GTPase activity of alpha-tubulin.

TUB3 also known as Tubulin Alpha 3 is a component of microtubules which are cylindrical structures within cells. It has a mass of approximately 50 kDa. TUB3 polymerizes with beta-tubulin to form the tubulin heterodimer which is the building block of microtubules. This protein is expressed in a variety of tissues including the brain and muscles which highlights its importance in these locations.
TUB3 plays a role in the stabilization of microtubule structures. As a part of the tubulin family it interacts within a larger complex to support cellular dynamics such as shape motility and division. It is essential for processes like intracellular transport where vesicles and organelles rely on microtubules to journey through the cytoplasm. TUB3 also contributes to the proper operation of the mitotic spindle during cell division ensuring accurate chromosome segregation.
Its involvement includes the cytoskeleton organization pathway and the mitosis pathway. During cell cycling TUB3 interacts with key proteins like kinesin and dynein which are responsible for motor activities along microtubules. This interaction supports mitotic spindle assembly an important process during the cell replication cycle. TUB3 facilitates various cellular mechanisms tied to structural maintenance and functional operations.
TUB3 has relevancy to neurodegenerative conditions and cancer. Mutations or dysregulation of TUB3 link to Alzheimer's disease where it might affect neuronal transport and stability. In cancer TUB3 expression levels can influence cell proliferation rates. The protein's interplay with many cancer-related kinases such as GSK-3 portrays its potential impact on tumor growth and advancement. Understanding and targeting TUB3 could offer new angles for therapeutic development in these areas.
