TUBA1A also known as alpha-tubulin 1A or Tubulin Alpha-1A Chain is a protein with a molecular mass of about 50 kDa. It is an important subunit of tubulin which polymerizes to form microtubules essential components of the cytoskeleton. TUBA1A is expressed in a wide range of tissues but shows higher levels in the central nervous system as tubulin forms the structural framework necessary for neuron function and cellular integrity.
Biological function summary

The TUBA1A protein functions in the assembly and disassembly of microtubules participating as part of the heterodimeric tubulin complex along with beta-tubulin. This structure allows microtubules to contribute to intracellular transport cell motility and chromosome segregation during cell division. The dynamic instability of microtubules facilitated by tubulin proteins like TUBA1A is important for the adaptive cellular functions.

Pathways

TUBA1A plays a role in the microtubule-associated processes being involved in the MAPK/ERK pathway and the mTOR signaling pathway. These pathways are important for cell growth differentiation and survival. TUBA1A's interaction with other microtubule-associated proteins regulates these signaling events and it also associates with proteins such as MAP2 or tau influencing the organization of microtubule networks.

Mutations in the TUBA1A gene relate to conditions like lissencephaly and microlissencephaly which affect brain development and lead to neurological deficits. In these disorders TUBA1A mutations disrupt normal microtubule function often influencing other proteins such as LIS1 which is critical for proper neuronal migration. Understanding TUBA1A's role can help in diagnosing and exploring therapeutics for such neurological conditions.

Tubulin is the major constituent of microtubules, a cylinder consisting of laterally associated linear protofilaments composed of alpha- and beta-tubulin heterodimers. Microtubules grow by the addition of GTP-tubulin dimers to the microtubule end, where a stabilizing cap forms. Below the cap, tubulin dimers are in GDP-bound state, owing to GTPase activity of alpha-tubulin.
