Tubulin is the major constituent of microtubules, a cylinder consisting of laterally associated linear protofilaments composed of alpha- and beta-tubulin heterodimers. Microtubules grow by the addition of GTP-tubulin dimers to the microtubule end, where a stabilizing cap forms. Below the cap, tubulin dimers are in GDP-bound state, owing to GTPase activity of alpha-tubulin.

TUBA8 also known as tubulin alpha 8 is a protein involved in maintaining the structure of microtubules. It has a molecular mass of about 50 kDa. This protein serves as an important component of the cytoskeleton helping in cellular shape and intracellular transport. Expression of TUBA8 occurs in various tissues including brain and testes but it is not restricted to these. TUBA8 forms heterodimers with beta-tubulin to construct microtubules playing an essential mechanical role in cell stability and movement.
TUBA8 contributes to important cellular processes including cell division movement and intracellular transport. It forms part of the microtubule network essential for the dynamic functions of the cytoskeleton. The protein interacts with other tubulin isoforms and various associated proteins facilitating proper cellular function and structure maintenance. By guiding vesicle transport TUBA8 further influences cell signaling and organization.
TUBA8 plays an important role in the microtubule assembly pathway and interacts with the mitogen-activated protein kinase (MAPK) pathway. TUBA8's proper functioning is necessary for cellular activities such as mitosis where microtubule dynamics are critical. In the MAPK pathway TUBA8 coordinates with proteins like MAPK1 and MAPK3 impacting cell survival and apoptosis. These interactions show TUBA8's importance in cellular response systems.
Abnormalities in TUBA8 are linked to complex disorders such as lissencephaly and microcephaly. These conditions involve disrupted brain development and abnormal nerve cell positioning highlighting TUBA8's role in neural development. Additionally changes in TUBA8 expression relate to disorders involving the protein LIS1 which also affects microtubule dynamics and brain development. Understanding TUBA8's contribution to these disorders aids in identifying potential therapeutic targets.
