Acts as accessory component of the N-oligosaccharyl transferase (OST) complex which catalyzes the transfer of a high mannose oligosaccharide from a lipid-linked oligosaccharide donor to an asparagine residue within an Asn-X-Ser/Thr consensus motif in nascent polypeptide chains. Involved in N-glycosylation of STT3B-dependent substrates. Specifically required for the glycosylation of a subset of acceptor sites that are near cysteine residues; in this function seems to act redundantly with MAGT1. In its oxidized form proposed to form transient mixed disulfides with a glycoprotein substrate to facilitate access of STT3B to the unmodified acceptor site. Has also oxidoreductase-independent functions in the STT3B-containing OST complex possibly involving substrate recognition.Magnesium transporter.

The protein involves itself in an important role in the cellular mechanism of N-glycosylation. TUSC3 associates with the oligosaccharyltransferase complex facilitating the transfer of oligosaccharides to nascent proteins. This process enhances the stability and function of many proteins that require glycosylation. TUSC3's proper activity ensures the correct folding and functionality of these glycoproteins impacting cell homeostasis and viability.
TUSC3 also known as Tumor Suppressor Candidate 3 is a protein weighing approximately 42 kDa. It plays a role in N-glycosylation an important post-translational modification mechanism. TUSC3 is mainly expressed in the human endoplasmic reticulum and is involved in protein folding processes. By aiding in this process TUSC3 supports the correct functioning of proteins throughout the cell. Within the endoplasmic reticulum it serves as part of a complex that ensures proper protein maturation.
TUSC3 functions within both the N-glycosylation pathway and the unfolded protein response (UPR). Its activity in N-glycosylation ensures proteins are correctly folded and functional while its role in the UPR helps manage cellular stress by promoting the proper folding of proteins under stress conditions. TUSC3 works closely with proteins like calnexin in these pathways collaborating to maintain protein quality control and reduce potential cellular dysfunction.
TUSC3 links to certain cancers and neurological conditions. Its role as a tumor suppressor implicates it in various cancers where reduced expression can lead to tumor progression. Additionally mutations or dysfunctions in TUSC3 have been associated with intellectual disabilities where glycosylation deficiencies contribute to disease pathology. TUSC3 interacts with proteins such as alpha-dystroglycan in these contexts where alterations can disrupt normal cellular functions and lead to disease development or progression.

Formalin-fixed, paraffin-embedded human colon cancer tissue stained for TUSC3 with ab230520 at a 1/100 dilution in immunohistochemical analysis.
The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with ab230520 at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with hematoxylin and mounted with DPX.
HeLa (Human epithelial cell line from cervix adenocarcinoma) cells stained for TUSC3 using ab230520 at a dilution of 1/100 in ICC/IF.
Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4°C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight® 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
Formalin-fixed, paraffin-embedded human testis tissue stained for TUSC3 with ab230520 at a 1/100 dilution in immunohistochemical analysis.
