HeLa (Human epithelial cell line from cervix adenocarcinoma) cells stained for TUSC3 using ab230520 at a dilution of 1/100 in ICC/IF.

Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4°C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight® 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

Formalin-fixed, paraffin-embedded human colon cancer tissue stained for TUSC3 with ab230520 at a 1/100 dilution in immunohistochemical analysis.

The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with ab230520 at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with hematoxylin and mounted with DPX.

TUSC3 also known as Tumor Suppressor Candidate 3 is a protein weighing approximately 42 kDa. It plays a role in N-glycosylation an important post-translational modification mechanism. TUSC3 is mainly expressed in the human endoplasmic reticulum and is involved in protein folding processes. By aiding in this process TUSC3 supports the correct functioning of proteins throughout the cell. Within the endoplasmic reticulum it serves as part of a complex that ensures proper protein maturation.
Biological function summary

The protein involves itself in an important role in the cellular mechanism of N-glycosylation. TUSC3 associates with the oligosaccharyltransferase complex facilitating the transfer of oligosaccharides to nascent proteins. Acts as accessory component of the N-oligosaccharyl transferase (OST) complex which catalyzes the transfer of a high mannose oligosaccharide from a lipid-linked oligosaccharide donor to an asparagine residue within an Asn-X-Ser/Thr consensus motif in nascent polypeptide chains. Involved in N-glycosylation of STT3B-dependent substrates. Specifically required for the glycosylation of a subset of acceptor sites that are near cysteine residues; in this function seems to act redundantly with MAGT1. In its oxidized form proposed to form transient mixed disulfides with a glycoprotein substrate to facilitate access of STT3B to the unmodified acceptor site. Also has oxidoreductase-independent functions in the STT3B-containing OST complex possibly involving substrate recognition. Could indirectly play a role in Mg(2+) transport (PubMed : 19717468).
