Target data

Function

Actin-binding protein involved in motile and morphological processes. Inhibits actin polymerization, likely by sequestering G-actin. By capping the barbed ends of filaments, it also regulates motility. Seems to play an important role in clathrin-mediated endocytosis and distribution of endocytic organelles (By similarity).

Activity summary

TWF1 also known as Twinfilin-1 or A6-related protein is an actin-binding protein with a molecular mass of about 38 kDa. This protein is expressed in various tissues including skeletal muscle heart and brain. TWF1 playing a role in actin dynamics binds to actin monomers and may affect actin filament turnover. It acts by promoting actin depolymerization and inhibiting actin nucleation contributing to the dynamic remodeling of the actin cytoskeleton.

Biological function summary

TWF1 is important in regulating cell motility morphology and division by influencing actin filament dynamics. TWF1 does not necessarily exist as part of a complex but its activity interacts with components of the cytoskeleton. By modulating actin dynamics TWF1 influences processes such as endocytosis cellular movement and response to external stimuli.

Pathways

Actin dynamics mediated by TWF1 is integral to the cytoskeleton organization pathway and the endocytic pathway. Within these pathways it interacts indirectly with proteins like cofilin and profilin which also regulate actin cytoskeleton. Its activity helps cells to adapt their shape and movement facilitating diverse cellular functions and signaling processes.

Associated diseases and disorders

TWF1 has relevance to cancer progression and conditions affecting cell mobility. Abnormal regulation of actin dynamics due to altered TWF1 activity might lead to enhanced cancer cell invasion and metastasis. Additionally TWF1 connects with proteins like ADF/Cofilin regarding cancer-related pathways influencing actin structure remodeling and cell movement in malignant cells.
