TWSG1 also known as TSG or Twisted Gastrulation Protein Homolog 1 is a secreted protein involved in the modulation of signaling pathways. It has a molecular mass of approximately 24 kDa. TWSG1 is expressed in a variety of tissues including the brain heart and skeletal muscle. This protein interacts directly with other molecules to influence cellular communication and development processes.
Biological function summary

TWSG1 interacts with members of the bone morphogenetic protein (BMP) family modulating their signaling activity. TWSG1 functions as part of a complex with other extracellular matrix proteins which helps regulate the availability and activity of BMPs. This regulation is essential during early embryonic development and tissue differentiation. By fine-tuning BMP signaling TWSG1 plays a significant role in proper developmental processes.

Pathways

TWSG1 is integral in the BMP signaling and TGF-beta pathways. These pathways are influential in promoting cell growth differentiation and apoptosis. In the BMP signaling pathway TWSG1 interacts with Noggin and Chordin. These interactions ensure balanced signaling which is necessary for correct tissue formation and organogenesis. The role of TWSG1 in regulating TGF-beta pathways further highlights its involvement in controlling cellular responses to environmental stimuli.

TWSG1 is linked to craniofacial malformations and cancer. Disruptions in BMP signaling where TWSG1 plays a regulatory role can lead to improper craniofacial development resulting in congenital deformities. Additionally aberrant expression of TWSG1 affects tumorigenic processes contributing to certain cancers. For example TWSG1 is connected to the overexpression of BMPs and TGF-beta proteins which can promote tumor growth and metastasis in various cancers.

May be involved in dorsoventral axis formation. Seems to antagonize BMP signaling by forming ternary complexes with CHRD and BMPs, thereby preventing BMPs from binding to their receptors. In addition to the anti-BMP function, also has pro-BMP activity, partly mediated by cleavage and degradation of CHRD, which releases BMPs from ternary complexes. May be an important modulator of BMP-regulated cartilage development and chondrocyte differentiation. May play a role in thymocyte development (By similarity).
