Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Tyrosine Hydroxylase antibody [EP1532Y] - Neuronal Marker – Goat IgG (Chimeric) (AB317785)
This image was produced using ab317795, the same antibody clone but in a different formulation. Immunofluorescence staining of Tyrosine Hydroxylase in sections of formalin-fixed paraffin-embedded rat brain (positive) and rat liver (negative). Performed on a Leica BOND. The section was pre-treated using heat mediated antigen retrieval with sodium citrate (pH6.0) for 20 minutes. The section was then incubated at room temperature for 1 hour with ab317795 at 1/100 dilution, and then incubated for 1 hour with ab150135 Donkey Anti-Goat IgG H&L (Alexa Fluor® 647) preadsorbed at 1/1000 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue). The section was then mounted using Dako Fluorescence Mounting Medium®. Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8). For other IHC staining systems (automated and non-automated), customers should optimize variable parameters such as antigen retrieval conditions, antibody concentrations and incubation times.

Tyrosine hydroxylase is essential for synthesizing catecholamines including dopamine norepinephrine and epinephrine. This enzyme associates with other components in the catecholamine biosynthetic pathway. It functions as part of a larger complex within certain cells where its action determines levels of important neurotransmitters. These neurotransmitters help regulate numerous central and peripheral nervous system activities.
Pathways
Tyrosine hydroxylase acts as a critical component in both the dopaminergic and adrenergic pathways. In the dopaminergic pathway tyrosine hydroxylase facilitates dopamine production influencing various physiological processes. The enzyme also correlates with ardrd protein in these pathways allowing fine-tuned control of neurotransmitter synthesis. Proteins related to tyrosine hydroxylase in these pathways include aromatic L-amino acid decarboxylase which further processes L-DOPA into dopamine.
