Supplementary information

TZFP also known as ZBTB32 is a protein encoded by the ZBTB32 gene. This protein weighs approximately 58 kDa and is a member of the BTB/POZ and zinc finger family of transcription factors. It acts as a transcriptional repressor by binding to specific DNA sequences and modulating gene expression. TZFP is expressed in various tissues with notable expression in lymphoid tissues and the thymus where it plays an important role in regulating immune cell function and development.
Biological function summary

TZFP/ZBTB32 influences cell proliferation and differentiation within the immune system. It is not known to be a part of a major protein complex but interacts directly with chromatin to regulate transcription. Through its action it affects immune responses by controlling the activation and repression of genes important for lymphocyte development and function. TZFP also plays roles in the modulation of immune tolerance and autoimmunity.

Pathways

TZFP/ZBTB32's function is evident in immune regulatory pathways. It interacts with signaling cascades like the NF-κB pathway where it serves as a modulator of transcriptional responses to inflammatory signals. Additionally TZFP engages with other transcription factors like BCL6 which are critical in B and T cell activities. This regulatory control helps ensure a balanced immune response necessary for maintaining homeostasis.

TZFP/ZBTB32 has been implicated in conditions involving immune dysregulation. For example aberrant expression of TZFP is associated with autoimmune diseases such as systemic lupus erythematosus (SLE). It may also relate to certain lymphomas where its altered expression can contribute to malignant transformation and progression. Through its involvement in these disorders proteins like NF-κB and BCL6 often interact influencing disease pathogenesis and offering potential therapeutic targets.

Target data

DNA-binding protein that binds to the to a 5'-TGTACAGTGT-3' core sequence. May function as a transcriptional transactivator and transcriptional repressor. Probably exerts its repressor effect by preventing GATA3 from binding to DNA. May play a role in regulating the differentiation and activation of helper T-cells (By similarity).
