U1A also known as snRNP U1A is a protein involved in the splicing of pre-messenger RNA. U1A is approximately 31 kDa in mass and functions as an important component of the U1 small nuclear ribonucleoprotein (snRNP) complex. Expressed in the cell nucleus U1A binds to the stem-loop structures of U1 RNA stabilizing the RNA-protein complex. This binding activity is critical as it allows U1 snRNP to recognize and bind splice sites on precursor mRNA facilitating the splicing process.
Biological function summary

U1A participates in the formation and function of the spliceosome a large RNA-protein complex essential for pre-mRNA processing. U1A as part of the U1 snRNP helps in recognizing and positioning the 5' splice site of introns in pre-mRNA. Through this role U1A ensures proper removal of non-coding sequences allowing for the maturation of mRNA. The spliceosome which includes other components like U2 U4 U5 and U6 snRNPs coordinates finely to execute the precise cutting and ligation of RNA sequences during splicing.

Pathways

U1A plays a central role in the mRNA splicing pathway which is a critical step in gene expression regulation. The splicing pathway interfaces with other biological pathways such as those involved in mRNA translation and degradation. U1A interacts with several proteins within these pathways notably SR proteins and hnRNPs which influence splice site selection and enhance the efficiency of splicing. These interactions highlight the interconnectedness of RNA processing events with overall gene expression controls.

U1A has been implicated in autoimmune diseases such as systemic lupus erythematosus (SLE). In SLE the body generates autoantibodies against several snRNP components including U1A which disrupts normal splicesome function. This leads to defective RNA processing and contributes to disease pathology. Additionally mutations or dysregulation of splicing proteins like SF3B1 and SRSF2 also associated with U1A through splicing abnormalities have been linked to various cancers where they lead to aberrant splicing patterns that drive tumorigenesis.

Component of the spliceosomal U1 snRNP, which is essential for recognition of the pre-mRNA 5' splice-site and the subsequent assembly of the spliceosome. U1 snRNP is the first snRNP to interact with pre-mRNA. This interaction is required for the subsequent binding of U2 snRNP and the U4/U6/U5 tri-snRNP. SNRPA binds stem loop II of U1 snRNA. In a snRNP-free form (SF-A) may be involved in coupled pre-mRNA splicing and polyadenylation process. May bind preferentially to the 5'-UGCAC-3' motif on RNAs.
