Uba6 also known as ubiquitin-like modifier activating enzyme 6 is an E1-like ubiquitin-activating enzyme. It has an approximate mass of around 120 kDa. This enzyme facilitates the activation of ubiquitin and similar proteins by catalyzing the formation of a ubiquitin-adenylate intermediate. Uba6 is expressed in a wide range of human tissues with higher levels observed in the brain heart and skeletal muscle. Its expression suggests a need for its activity in diverse biological processes across different tissue types.
Biological function summary

Uba6 modifies proteins by attaching ubiquitin moieties which tags them for various fates including degradation or location changes. Uba6 functions as a part of the ubiquitin-proteasome system (UPS) a critical protein degradation and quality control pathway. In this system Uba6 collaborates with E2 conjugating enzymes such as Ube2z which transfer ubiquitin from Uba6 to substrate proteins. This involvement emphasizes its role in protein homeostasis and cellular stress responses by controlling protein stability.

Activates ubiquitin by first adenylating its C-terminal glycine residue with ATP, and thereafter linking this residue to the side chain of a cysteine residue in E1, yielding a ubiquitin-E1 thioester and free AMP. Specific for ubiquitin, does not activate ubiquitin-like peptides. Differs from UBE1 in its specificity for substrate E2 charging. Does not charge cell cycle E2s, such as CDC34. Essential for embryonic development. Required for UBD/FAT10 conjugation. Isoform 2 may play a key role in ubiquitin system and may influence spermatogenesis and male fertility.
