Supplementary information

UBAP2L also known as USP10-associated protein 2-like has a molecular mass of approximately 115 kDa. Mechanically it functions as a translational regulator. This protein is enriched in cellular compartments including the cytoplasm and stress granules. It interacts with multiple proteins and is found in many tissues suggesting a role in cellular stress responses and phase separation events.
Biological function summary

UBAP2L influences stress granule dynamics and protein synthesis under stress conditions. It is important for assembly of these translationally silent granules suggesting its involvement in cell survival. As a part of a larger protein interaction complex UBAP2L contributes to cellular homeostasis. Its interaction partners include G3BP1 which serves as an important player in stress granule nucleation.

Pathways

UBAP2L integrates into the mRNA surveillance pathway and the cellular stress response pathway. It interacts with proteins like USP10 and G3BP1 to facilitate stress granule formation and disassembly critical for proper mRNA metabolism. Through these pathways UBAP2L helps maintain proteostasis under cellular stress affecting translation and decay of mRNA.

UBAP2L has implications in neurodegenerative diseases and cancer. Aberrations in its function contribute to Alzheimer's disease through dysregulated stress granule assembly involving proteins like TDP-43. Additionally it plays a role in tumor progression where alterations in its expression correlate with poor prognosis interacting with oncogenic proteins such as HNRNPA1 linked to metastasis and cellular proliferation.

Target data

Recruits the ubiquitination machinery to RNA polymerase II for polyubiquitination, removal and degradation, when the transcription-coupled nucleotide excision repair (TC-NER) machinery fails to resolve DNA damage (PubMed : 35633597). Plays an important role in the activity of long-term repopulating hematopoietic stem cells (LT-HSCs) (By similarity). Is a regulator of stress granule assembly, required for their efficient formation (PubMed : 29395067, PubMed : 35977029). Required for proper brain development and neocortex lamination (By similarity).
