UBASH3A also known as suppressor of T-cell receptor signaling 1 (STS-1) is a protein involved in regulating T-cell signaling. It has a molecular mass of approximately 67 kDa. UBASH3A is expressed mainly in lymphoid tissues including the thymus and spleen. The protein contains a Src homology 3 (SH3) domain and a ubiquitin-associated (UBA) domain which are important for its interactions with other proteins and its function in cellular signaling processes.
Biological function summary

UBASH3A plays an important role in modulating immune responses. It regulates T-cell receptor (TCR) signaling by acting as a negative regulator ensuring proper immune tolerance and preventing overstimulation of immune responses. UBASH3A is not known to be part of a larger protein complex. It interacts with signaling molecules and adaptor proteins to modulate signal transduction pathways in T-cells controlling their activation and proliferation.

Pathways

UBASH3A influences T-cell signaling pathways and the broader immune response. It is involved in the TCR signaling pathway which is essential for the activation and development of T-cells. Through this pathway UBASH3A interacts with proteins like LCK and ZAP-70 which are necessary for TCR-induced signaling events leading to cytokine production and cell-mediated immune responses. Additionally UBASH3A is connected to pathways controlling apoptosis where it fine-tunes the balance between cell survival and programmed cell death.

UBASH3A has implications in autoimmune diseases and cancer. Its role as a negative regulator of TCR signaling links it to autoimmune conditions such as type 1 diabetes where improper TCR signaling can lead to autoimmune attack of pancreatic beta cells. Furthermore altered expression or function of UBASH3A can impact cancer progression particularly in hematological cancers like leukemia where T-cell signaling dysregulation can promote proliferation. Proteins such as CTLA-4 and PD-1 which are involved in immune checkpoint pathways can be indirectly influenced by UBASH3A activity affecting disease progression and treatment outcomes.

Interferes with CBL-mediated down-regulation and degradation of receptor-type tyrosine kinases. Promotes accumulation of activated target receptors, such as T-cell receptors, EGFR and PDGFRB, on the cell surface. Exhibits negligible protein tyrosine phosphatase activity at neutral pH. May act as a dominant-negative regulator of UBASH3B-dependent dephosphorylation. May inhibit dynamin-dependent endocytic pathways by functionally sequestering dynamin via its SH3 domain.
