Supplementary information

UBE2Z also known as Ubiquitin-Conjugating Enzyme E2 Z is an enzyme that plays a mechanical role in the ubiquitination process. This enzyme belongs to the E2 ubiquitin-conjugating enzyme family. UBE2Z has a mass of approximately 37.5 kDa. It is expressed in various tissues including but not limited to the liver heart and skeletal muscle. The enzyme catalyzes the transfer of ubiquitin from an E1 ubiquitin-activating enzyme to E3 ubiquitin ligases facilitating substrate ubiquitination.
Biological function summary

UBE2Z is involved in protein degradation and regulation of protein homeostasis. It is an important part of the ubiquitin-proteasome system ensuring damaged or misfolded proteins are marked for degradation. UBE2Z can function independently or in conjunction with multi-protein complexes. It has an important role in regulating cellular processes like cell cycle progression DNA repair and response to stress. Additionally it contributes to the regulation of several cellular pathways through these processes.

Pathways

UBE2Z is significantly involved in the protein ubiquitination pathway and cellular response to oxidative stress. In the protein ubiquitination pathway it interacts with various E3 ligases influencing protein turnover. Notably it interacts with the KEAP1 pathway which regulates the degradation of NRF2 a transcription factor involved in antioxidative responses. UBE2Z’s association with these pathways links it to the modulation of protein stability and cellular sensitivity to oxidative damage.

UBE2Z has been linked to metabolic disorders and renal diseases. Research has shown that variations in the UBE2Z gene have associations with obesity-related traits and Type 2 diabetes. The enzyme also has connections to proteins like NRF2 and KEAP1 which play roles in the development of renal pathologies. Dysregulation in UBE2Z activity can contribute to oxidative stress and inflammation further influencing disease progression in these disorders.
