Accepts ubiquitin from the E2 ubiquitin-conjugating enzyme UBE2D1 in the form of a thioester and then directly transfers the ubiquitin to targeted substrates (PubMed:32039437, PubMed:9575161). Associates with the proteasome and promotes elongation of ubiquitin chains on substrates bound to the 26S proteasome (PubMed:24158444, PubMed:28396413, PubMed:31375563). Also catalyzes 'Lys-29'- and 'Lys-48'-linked ubiquitination of 26S proteasome subunit ADRM1/RPN13 in response to proteotoxic stress, impairing the ability of the proteasome to bind and degrade ubiquitin-conjugated proteins (PubMed:24811749, PubMed:31375563). Acts as a negative regulator of autophagy by mediating 'Lys-29'- and 'Lys-48'-linked ubiquitination of PIK3C3/VPS34, promoting its degradation (PubMed:33637724). Can assemble unanchored poly-ubiquitin chains in either 'Lys-29'- or 'Lys-48'-linked polyubiquitin chains; with some preference for 'Lys-48' linkages (PubMed:11278995, PubMed:16601690, PubMed:25752577). Acts as a negative regulator of type I interferon by mediating 'Lys-48'-linked ubiquitination of IRF3 and IRF7, leading to their degradation by the proteasome (PubMed:21167755). ab226173 was affinity purified using an epitope specific to UBE3C immobilized on solid support.
UBE3C also known as ubiquitin protein ligase E3C is an E3 ubiquitin-protein ligase that plays a mechanical role in tagging proteins for degradation. This tagging involves attaching ubiquitin molecules to substrate proteins which targets them for proteasomal degradation. UBE3C has a molecular mass of approximately 90 kDa. Expression of this protein occurs in various tissues with higher levels found in the brain and testis suggesting these organs may rely more on its function.
UBE3C regulates protein homeostasis by working as part of the ubiquitin-proteasome system which maintains cellular function by removing damaged or unneeded proteins. It assists in controlling protein quality and stability. Although not typically part of larger multiprotein complexes UBE3C influences the turnover of several cellular proteins. Its action affects many processes including neuronal function and response to cellular stress.
UBE3C activity intersects with the ubiquitin-proteasome pathway a fundamental mechanism for protein degradation and turnover in cells. This pathway ensures cellular protein quality and prevents toxic protein accumulation. It has associations with proteins like UCHL1 which also have roles in the ubiquitination process. Moreover UBE3C links to the Wnt signaling pathway by modulating the stability of pathway components such as β-catenin which influences cell proliferation and differentiation.
UBE3C involvement appears significant in neurodegenerative diseases and certain cancers. In neurodegenerative conditions like Alzheimer's disease altered UBE3C expression or function might impact protein aggregates formation similar to its interaction with proteins like tau. In cancer abnormal UBE3C levels relate to changes in protein degradation that can lead to unchecked cell growth and survival affecting proteins like p53 an important tumor suppressor.
