Supplementary information

UBFD1 also known as Ubiquitin Family Domain-Containing Protein 1 is a protein with a molecular mass of approximately 35 kDa. It functions mechanically by interacting with ubiquitin tags playing a role in protein degradation and localization through the ubiquitin-proteasome system. UBFD1 is expressed in various tissues with notable expression in the liver brain and immune cells. This widespread expression suggests its involvement in multiple cellular functions.
Biological function summary

UBFD1 engages in cellular homeostasis by influencing protein turnover and signaling pathways. It is not known as part of a traditional protein complex but it interacts with several ubiquitin-associated proteins. UBFD1 regulates the stability of proteins by mediating their ubiquitination status which affects the cell cycle DNA repair and response to stress conditions. Its role is important for maintaining normal cellular function and preventing the accumulation of damaged proteins.

Pathways

UBFD1 influences many cellular processes notably the ubiquitin-proteasome pathway and the DNA damage response pathway. It works closely with proteins such as ubiquitin-conjugating enzymes and substrate-specific ubiquitin ligases. These pathways are integral to maintaining protein homeostasis and ensuring proper cellular responses to damage and stress. By modulating these pathways UBFD1 ensures effective protein management and repair mechanisms within the cell.

UBFD1 is linked to neurodegenerative diseases and certain types of cancer. Abnormal regulation or mutations in UBFD1 may contribute to protein misfolding and aggregation seen in diseases like Alzheimer's. In cancer UBFD1 dysfunction may lead to unregulated cell division due to impaired protein degradation. Studies have shown interactions between UBFD1 and proteins such as p53 highlighting its relevance in cancer pathogenesis through disruption of normal cell cycle control.
