UCHL4 also known as Ubiquitin C-terminal hydrolase L4 is a deubiquitinating enzyme with a mass of approximately 63 kDa. This protein is part of the ubiquitin-proteasome system which is vital in regulating protein degradation and turnover inside cells. UCHL4 is widely expressed in various tissues including the brain where it is involved in the processing of ubiquitinated proteins. As a hydrolase UCHL4 specifically removes ubiquitin from ubiquitinated protein substrates impacting protein stability and activity.
Deubiquitinating enzymes like UCHL4 play a role in maintaining cellular homeostasis by regulating the levels of proteins targeted for degradation. UCHL4 associates with other proteins to form complexes involved in the recycling of ubiquitin a process essential for protein quality control. This function makes UCHL4 an important regulator in various cellular processes such as cell cycle progression and DNA repair mechanisms. By removing ubiquitin UCHL4 helps in rescuing proteins from degradation and can influence cellular signaling pathways.
UCHL4 is involved in the ubiquitin-proteasome pathway (UPP) as well as the endosomal sorting complex required for transport (ESCRT). In the UPP UCHL4 works alongside proteins like the proteasome subunit beta type-7 (PSMB7) to mediate protein recycling and degradation. This pathway is essential for maintaining protein homeostasis within cells. In the ESCRT pathway UCHL4 assists in the sorting and trafficking processes critical for membrane dynamics and signaling.
