Western blot : Anti-UCP1 antibody (ab23841) staining at 1 ug/ml, shown in green; Mouse anti-Alpha Tubulin [DM1A] (ab7291) loading control staining at 1/20000 dilution, shown in magenta. In Western blot, ab23841 was shown to bind specifically to UCP1. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit IgG H&L 800CW and Goat anti-Mouse IgG H&L 680RD at 1/20000 dilution.

Uncoupling Protein 1 (UCP1) also known as thermogenin is an important protein found in the mitochondria of brown adipose tissue (BAT). It has a molecular mass of approximately 33 kDa. UCP1 functions as a mitochondrial carrier specifically involved in the process of proton transport across the inner mitochondrial membrane. It leads to the dissipation of the proton gradient producing heat instead of ATP. This process is called non-shivering thermogenesis. UCP1 is mainly expressed in brown adipose tissue an area important for heat production in mammals.
Biological function summary

UCP1 plays an essential role in energy metabolism by activating thermogenesis in brown adipocytes. It does not form a larger multi-protein complex but acts as an independent protein facilitating a significant amount of heat production. By uncoupling oxidative phosphorylation UCP1 controls energy expenditure and is central to maintaining body temperature in cold environments.
