UCP3 + UCP1 were immunoprecipitated from 1mg of Mouse skeletal muscle whole cell lysate with ab193470 at 1/50 dilution. Western blot was performed from the immunoprecipitate using ab193470 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/1000 dilution.

UCP3 also known as Uncoupling protein 3 is a mitochondrial protein that plays a role in the process of thermogenesis by dissipating the proton gradient. It is a member of the mitochondrial anion carrier proteins (MACP) and has a molecular mass of approximately 33 kDa. UCP3 is mostly expressed in skeletal muscle and to a lesser extent in brown adipose tissue. Its homolog UCP1 sometimes referred to as thermogenin has a similar function in brown adipose tissue where it participates prominently in heat production.
Biological function summary

Uncoupling proteins like UCP3 facilitate the regulation of energy balance and metabolism. They act by uncoupling ATP synthesis from the electrochemical gradient therefore reducing reactive oxygen species production. UCP3 is not part of a protein complex but interacts closely with the mitochondrial environment. UCP1 on the other hand is active in non-shivering thermogenesis within brown fat cells enabling organisms to fend off hypothermia.

Pathways

These uncoupling proteins play roles in energy homeostasis and lipid metabolism. UCP3 participates significantly in the fatty acid oxidation pathway influencing how cells utilize fatty acids for energy. UCP1 is an important player in the mitochondrial oxidative phosphorylation pathway where it influences heat production and energy expenditure. They are both connected to the broader metabolic pathways essential for maintaining energy balance interacting with other mitochondrial carriers.

UCP3 and UCP1 are associated with conditions like obesity and type 2 diabetes. UCP3 helps protect against lipid-induced mitochondrial damage which can be a factor in insulin resistance. Meanwhile UCP1’s disruption could lead to decreased ability in non-shivering thermogenesis contributing to weight gain. Both proteins hold therapeutic promise as targets to modulate metabolic conditions by influencing the body's energy expenditure.

Mitochondrial protein responsible for thermogenic respiration, a specialized capacity of brown adipose tissue and beige fat that participates in non-shivering adaptive thermogenesis to temperature and diet variations and more generally to the regulation of energy balance (By similarity). Functions as a long-chain fatty acid/LCFA and proton symporter, simultaneously transporting one LCFA and one proton through the inner mitochondrial membrane (PubMed : 24196960, PubMed : 28781081). However, LCFAs remaining associated with the transporter via their hydrophobic tails, it results in an apparent transport of protons activated by LCFAs. Thereby, dissipates the mitochondrial proton gradient and converts the energy of substrate oxydation into heat instead of ATP.
